New doubts about prostate-cancer vaccine Provenge | Health ...


NEW YORK (Reuters) ? Prostate cancer vaccine Provenge has long incited passions unlike any other cancer therapy.

Doctors who raised doubts about it received death threats. Health regulators and lawmakers faced loud protests at their offices. A physician at the American Cancer Society was so intimidated by Provenge partisans that he yanked a skeptical discussion of it from his blog.

The vitriol dissipated in April 2010, when the U.S. Food and Drug Administration approved Provenge for advanced prostate cancer, satisfying investors in manufacturer Dendreon and patients who for years had demanded it be put on the market.

But the bell on Round Two sounded when Marie Huber, a trained scientist and former hedge-fund analyst, made it her mission in the last year to analyze what she believes are deadly flaws in the studies that led to the approval of Provenge by the FDA.

She argues that the main reason Provenge seemed to extend survival ? a crucial factor in the FDA?s decision ? was that older men in the study who did not receive Provenge died months sooner than similar patients in other studies.

She raises the possibility the ?placebo? they received was actually harmful and made Provenge, known scientifically as sipuleucel-T, look better by comparison.

As Huber gains traction, most notably with a February paper in the prestigious Journal of the National Cancer Institute, she, too, is receiving threats. One post on an investors? message board last month suggested that ?somebody smack her with a rubber hose.? An email said ?don?t think you will be unscathed in this battle you waged on Provenge.?

Provenge is Dendreon?s only product and the company?s stature with investors has waned with disappointing sales. In 2011, product revenues totaled $213.5 million, far from the $400 million Dendreon initially projected.

The company insists Huber?s analysis is flawed and that Provenge has helped thousands of men with prostate cancer.

?I?m looking forward to getting this to patients around the world,? said President and Chief Executive John Johnson.

FIRST CANCER VACCINE

Since it won FDA approval two years ago, Provenge has been Exhibit A for the idea that a patient?s immune system can control or cure cancer. The first therapeutic cancer vaccine to reach the market, Provenge tries to engineer white blood cells, part of the immune system, to vanquish prostate cancer, which killed an estimated 33,720 men in the United States last year.

Its path to approval has all the features of a heavyweight healthcare fight ? desperate patients demanding access to a promising therapy, a very expensive drug that extends life only a few months and efficacy data open to interpretation.

The FDA declined to approve the drug in 2007, when a clinical trial failed to show it slowed tumor growth. That incited protests, lawsuits and death threats against physicians on the FDA advisory panel who did not recommend approval, breaking with the 13-4 majority in favor.

?Provenge came along when we didn?t have much to offer for prostate cancer,? said Dr. Len Lichtenfeld of the ACS. ?The advocacy community was bursting at the seams for something that worked. When you have that situation, it inflames passions and that can overtake the science.?

In the pivotal trial called IMPACT, published in July 2010 but shared with the FDA months earlier, Provenge extended median survival by 4.1 months to 25.8 months from 21.7 months. That was sufficient for FDA approval. The vaccine costs $93,000 and patients also incur physician and other charges. Medicare agreed to cover Provenge last year, as have private insurers, but doctors initially balked at a long wait for reimbursement.

Huber had long been ?utterly intrigued? by Provenge and its ?huge promise of harnessing the immune system to battle cancer,? she said in an interview.

In documents JNCI requires authors to sign, she declared no financial conflicts of interest. Neither she nor her former firm nor anyone else she is connected to stands to benefit financially from her analysis, she said.

Instead, she says she is motivated to help ?vulnerable and desperate patients? ? so much so that she gave up her job, salary and health insurance. Arguing that Provenge is harming these men, she called ?the whole thing utterly horrific. The company got away with hiding data and doctors making $7,000 per prescription won?t even engage in discussion? about whether it helps their patients.

After receiving degrees in biochemistry and bioscience enterprise from Cambridge University, Huber began working as an analyst for a hedge fund in 2007. A Thomson Reuters analysis of securities filings confirmed her former firm has not held any positions in Dendreon.

LACK OF EVIDENCE

Each dose of Provenge is custom-made. A nurse or technician withdraws white blood cells from a man?s arm in a three-to-four hour procedure called leukapheresis.

The cells are shipped to a Dendreon manufacturing facility, where for two days they are incubated with a ?fusion protein?: One protein that stimulates the cells? growth and maturation and another called PAP, or prostatic acid phosphatase. PAP is an antigen that studs prostate cancer cells like antennae, pieces of it sticking out of the cells? surfaces.

Dendreon says the patients? white blood cells take up the antigen and within hours their surfaces bristle with fragments of the telltale molecule. The cells are then shipped back to the physician and infused into the patient. A full treatment includes three such procedures, two weeks apart.

Back inside the body, Dendreon claims the modified cells trigger the immune system to produce T cells that kill any cell sporting the PAP antigen ? namely, prostate cancer cells.

In principle, that should eliminate the cancer, but Provenge does not shrink either the primary tumor or metastases.

Steven Rosenberg of the National Cancer Institute, a leading tumor immunologist, says that raises doubts over whether Provenge helps patients live longer, as the IMPACT trial reported.

?We have a lot of data that supports the idea that the product works the way it was designed to,? said Dr. Mark Frohlich, Dendreon?s chief medical officer. ?We?re seeing evidence of immune-system activation. The only question is whether the T cells are killing the tumor.?

The FDA acknowledges that data supporting Provenge?s approval did not show the drug shrank tumors, but says the overall survival benefit was enough to bring it to market. Spokeswoman Rita Chapelle, citing data submitted by Dendreon, said there is a ?lack of evidence of anti-tumor activity,? the reason for which ?is unclear.?

SUSPICIONS OVER SURVIVAL BENEFIT

Huber?s analysis comes from data showing that men who received the placebo had very different survival times based on their age. Men older than 65 lived 17.3 months on placebo and 23 months with Provenge. Men younger than 65 lived 28 months after receiving placebo and 29 months after Provenge.

Other studies have shown that age generally does not affect how long a man survives with this form of prostate cancer, says Peter Iversen, a urologist and prostate-cancer surgeon at the University of Copenhagen and co-author of the paper with Huber.

Combining these findings led to the new paper?s conclusion: The four-month edge in median survival from Provenge for all patients was due to longer survival among older men who got the vaccine.

?There is no efficacy in the younger patients, the primary group where you would expect it,? said Huber.

Since the immune system weakens with age, an immune-based therapy should work better in younger men.

Some experts agree.

?If it was really a vaccine, you?d think younger men would show more response, since they are more immunocompetent,? said NCI?s Rosenberg.

On that basis, Huber and her co-authors, including two prostate-cancer specialists, argue the placebo used in the trial may have harmed the older men, cutting months off their lives and inadvertently making Provenge seem beneficial.

One way that could have occurred was through leukapheresis. That process removed about 90 percent of certain kinds of circulating white blood cells, according to calculations by immunologist Laura Haynes of the Trudeau Institute, a co-author of the JNCI paper.

The Provenge men got back about 32 percent of those cells, which had been stored at body temperature. The placebo men got back 12 percent, which had been incubated at near-freezing temperatures. Cold storage has been reported to kill ?most, if not all, of those cells,? notes the JNCI paper. Moreover, said Haynes, ?if you return dead and dying cells to older men you are likely to cause inflammation,? which can stoke the growth of cancerous cells.

Younger men were better able to replace the lost white blood cells, argued Iversen. Older men could not, resulting in early death.

?These cells are very specialized and there is research suggesting that removing them can harm older men,? he said.

Earlier this month, DynaMed, an online database used by physicians, added to its Provenge entry a note on the JNCI paper, but calls the concern ?not substantiated.? ACS?s Lichtenfeld says the analysis ?might inhibit some patients and doctors from going ahead with a very expensive drug.?

Huber says she plans to approach European regulators as they consider Dendreon?s application to approve Provenge.

Investor message boards have lit up in response to the new paper. In February, an anonymous commentator on InvestorVillage.com warned that Huber?s work was about to be published ?a few days before our earnings. Her agenda is obvious.?

IFFY STATISTICS

Critics of the new analysis argue the number of cells removed is too small to suppress the immune system. Charles Drake, an oncologist and immunologist at Johns Hopkins Sidney Kimmel Comprehensive Cancer Center, said there is no evidence the placebo men in IMPACT suffered more infections or other effects of a depleted immune system than the Provenge men.

The scientist who led IMPACT, oncologist Philip Kantoff of Dana Farber Cancer Center, said colleagues in immunology ?dismissed as nonsense the idea that leukapheresis could hurt individuals.?

He takes issue, too, with the statistics. Dividing the men by whether they are older or younger than 65, he said, is ?arbitrary? and to pick apart data retrospectively is a statistical no-no.

Dendreon?s Frohlich also criticizes the statistics: ?If you do enough of these (post-hoc analyses) then by chance alone you?d expect to get one positive finding.?

In other words, it is almost always possible to find a subset of patients who do better than others.

When Dendreon divided the men by whether they were older or younger than about 71, he added, they found no red flags.

The FDA agrees that such post-hoc statistical analyses ?are exploratory? and their results ?must be interpreted with caution, as acknowledged by the authors.? Yet a paid consultant to Dendreon before the IMPACT trial agreed with many of Huber?s concerns.

?The control vaccine used in IMPACT and in the predecessor trial had never been used anywhere for anything and may well have been detrimental to patients,? said Donald Berry of MD Anderson Cancer Center, a leading biostatistician. ?Here?s a great way to get your drug approved: Kill the control patients.?

Despite the heated rhetoric, Provenge may go out with a whimper more than a bang. Promising new agents for advanced prostate cancer include an oral drug from Medivation Inc called enzalutamide, in the final phase of clinical trials, and Zytiga from Johnson Johnson, which won FDA approval in 2011. A vaccine that targets PSA, from Bavarian Nordic Immunotherapy, is in late-stage trials.

Dendreon does not disclose how many patients have been prescribed Provenge. CEO Johnson said that about 70 percent of its Provenge revenue comes from sales to community hospitals and doctors and 30 percent from academic medical centers. Some of the latter decline to use Provenge, deterred by lingering concerns over whether it provides a meaningful benefit.

Three such facilities in the Midwest, contacted at random by Reuters, confirmed they do not recommend Provenge. All asked not to be named for fear of receiving threats.

?It is my policy not to make public comments about this drug,? said one oncologist. Patients who ask for it ?are referred to another facility.?

Milky Way image reveals detail of a billion stars

ScienceDaily (Mar. 28, 2012) ? More than one billion stars in the Milky Way can be seen together in detail for the first time in an image captured by an international team of astronomers. Scientists created the colour picture by combining infra-red light images from telescopes in the northern and southern hemispheres. Large structures of the Milky Way galaxy, such as gas and dust clouds where stars have formed and died, can be seen in the image.

Dr Nick Cross of the University of Edinburgh will present the new work on March 29 at the National Astronomy Meeting in Manchester.

The picture represents part of a 10-year project involving scientists from the UK, Europe and Chile, who gathered data from the two telescopes. The information has been processed and archived by teams at the Universities of Edinburgh and Cambridge, who have made it available to astronomers around the world for further studies.

Archived information from the project -- known as the VISTA Data Flow System -- is expected to enable scientists to carry out groundbreaking research in future years without the need to generate further data.

The image shows the plane of the Milky Way galaxy, which is often described as looking like two fried eggs back-to-back, with a flat disc in the middle. Earth is close to the edge of this disc, and the image shows a cross-section through the disc as seen from Earth's perspective.

It combines data from the UKIDSS/GPS sky survey taken by the UK Infrared Telescope in Hawaii with the VVV survey from the VISTA telescope in Chile.

Astronomers used infra-red radiation instead of visible light to enable them to see through much of the dust in the Milky Way and record details of the centre of the galaxy.

The work was supported by the Science and Technology Facilities Council.

Dr Nick Cross, of the University of Edinburgh's School of Physics and Astronomy, said: "This incredible image gives us a new perspective of our galaxy, and illustrates the far-reaching discoveries we can make from large sky surveys. Having data processed, archived and published by dedicated teams leaves other scientists free to concentrate on using the data, and is a very cost-effective way to do astronomy."

Focus now on insurance renewal for 2013 ? Business Management ...

For most U.S. employers, open enrollment for health insurance benefits came and went by Jan. 1. And if you?re like plenty of comp and benefits professionals, the experience left you ex??hausted and glad to be done with it.

Too bad. Now is the time to start planning for the next insurance renewal go-round. An early start will give you a fighting chance to keep costs under control in 2013.

Start by reviewing how your most recent renewal went.

No plan? Plan to fail

If you?re going to get creative about combating rising health insurance costs, you need a well-thought-out strategic plan.

It?s not enough to wait for the usual renewal offer from your carrier next fall?typically 60 days before your plan expires. Realistically, that gives you no time to explore alternatives, either from your existing plan or a competitor.

As you plan for next year, ask these questions:

? What problems did you have with last year?s renewal?

Make a list while it?s still fresh in your mind. Did you have enough time to fully explore alternative plan de??signs? Alternative funding arrangements? Alternative carriers? If you wait until the fall to focus on those issues, you?re more likely to confront the same problems you had before.

? Does your renewal date fit your business plan?

Would it make more sense to time renewal to coincide with your fiscal year or some other cycle?

For many companies, the fourth quarter is the busiest time of the year. That can make it hard to communicate ben??efits changes to distracted employees?and get them to turn in their paperwork.

Added benefit: Get ready for better service. Tying your renewal to the new year means you?re competing with al??most every other employer for the attention of brokers and insurance carriers.

? What were the rates of the alternative plans presented the past two renewals?

At each renewal, most carriers provide several alternative plans in addition to your current plan. Refer back to your old renewals to begin the process of estimating the cost of migrating your employees to alternative coverage.

If your broker or carrier presented the same or similar alternatives the past two years, you?ll be able to see how those plans are trending compared to yours.

? How do your retention and turnover figures compare to your goals?

If you conduct exit interviews, have employees mentioned dissatisfaction with health benefits as a reason for leaving? Your next plan may be able to address those concerns.

Ask your broker or carrier to prepare total compensation statements to educate employees on the value of company-provided benefits.

Tip: If you can, check with HR colleagues in other organizations. If they have better retention numbers, find out if better benefits are enticing employees to stay. If so, ask for a referral to a new broker or carrier.

? How much of an increase for future renewals can you afford?

Here?s where you need to be tuned into your organization?s overall fiscal plan. Engage leaders now in a real conversation about how much they are willing to commit to benefits.

? Can your next plan lower costs by helping employees stay healthy?

Disease-management programs can help prevent a catastrophic claim from developing. But what about implementing a wellness program to prevent chronic conditions from manifesting in the first place? Waiting until an employee or a dependent becomes a diabetic and then figuring out how to manage those claims invariably means higher costs for everyone in the plan.

Now is the time to explore adding a wellness program to your plan.

(AP) ? The discovery of a body believed to be that of a beloved teacher at a prestigious boarding school sent shudders of grief and anxiety through a small New England town a day after her SUV was found running with her unharmed 2-year-old boy inside.

Melissa Jenkins, a 33-year-old single mother, taught science at St. Johnsbury Academy, a school of about 970 students that was established in the 1840s and whose alumni include former President Calvin Coolidge.

Vermont State Police Maj. Ed Ledo would not give details on the condition of the body found Monday in Barnet, a town not far St. Johnsbury. A friend who was looking for Jenkins called police Sunday and Jenkins' vehicle was discovered that evening near signs of a struggle.

An autopsy was planned for Tuesday. A family friend is caring for the boy. His father, B.J. Robertson, would not comment on Jenkins' disappearance.

Authorities acknowledged they did not know whether Jenkins' disappearance was isolated, and Ledo said at a news conference that the public should be vigilant as authorities continue to seek a suspect.

Throughout the day, townsfolk converged at the restaurant where Jenkins worked part-time, seeking solace and updates. As they braved bone-chilling winds for an evening candlelight service, news about the discovery of the body began filtering through the crowd.

"She would do anything for anybody. She definitely will be greatly missed," said Ron Craig, of Peacham, who said he and his wife occasionally babysat Jenkins' son.

Craig said it's scary that police did not know if this was an isolated incident. "We've been locking our doors all the time because you just don't know what's going to happen," he said.

Jenkins' vehicle was found not far from her home in a rural area at 11:30 p.m. Sunday. She had no restraining orders out on anyone, police said.

Jenkins was a girls freshman basketball coach and a dorm proctor until she had her son. She graduated from Lyndon State College with a degree in natural science and geology. She was working on her master's degree, headmaster Tom Lovett said.

"She's got a real gift with students who either haven't liked science before or learning science doesn't come easy to them," Lovett said. "She's got a real gift with them."

St. Johnsbury Academy also serves as a public school for the town, about 40 miles south of the Canadian border.

Jenkins was a waitress at night at The Creamery Restaurant in Danville, the eatery where co-workers, friends and the father of Jenkins' son gathered Monday afternoon along with others who were curious or concerned.

"We all know her. It's a tough thing right now," said Marion Cairns, the owner, who described Jenkins as bright, pretty, a good mother and fun to be around. "She'd cut her arms off before she'd let anybody touch that boy. I mean, that boy meant everything to her."

Eric Berry, 44, of Lyndonville, a cousin by marriage whose daughter is Jenkins' goddaughter, described her as a beautiful, kind person whom he believes was coming to someone's aid when she disappeared.

"She left her house with the idea, I think, to try to help somebody, and that's as far as I'm going to go with that because I don't want to damage any investigation," he said.

The academy will provide counseling to grieving students, Lovett said.

The disappearance recalled that of 20-year-old Krista Dittmeyer of Portland, Maine, whose car was found idling with its hazard lights on, her 14-month-old daughter unharmed, a year ago about 50 miles away in New Hampshire. Dittmeyer's body was found in a pond. Three men were arrested on charges in her robbery and killing.

Authorities said Monday there is no indication the cases were related.

Belfast: reluctant birthplace of the Titanic

The city tried to ignore the Titanic until recently because of guilt over its loss, but is now celebrating the shipbuilding genius of Belfast workers at the new Belfast Titanic museum.

?Belfast was a byword for gloom during the Troubles that spanned from the late 1960s to late 1990s, but it is experiencing a new vitality.

Ironically this coincides with the anniversary of the tragedy of the RMS Titanic, which sank 100 years ago on April 15 with a loss of more than 1,500 lives. Belfast is set to commemorate this sad milestone with church services and other events.

The city, which tried to ignore the Titanic until recently because of misplaced guilt at her loss, is now celebrating the shipbuilding genius of the Belfast workers who made her the most advanced vessel in the world at the time. Two recent developments helped the Titanic to become a source of pride: the discovery of the wreck in 1985 by Robert Ballard and the 1997 blockbuster movie, ?Titanic.?

Gradually it became respectable to talk about the Titanic in Belfast, and the city?s shipbuilding history will be celebrated during the Titanic Belfast Festival in April.

The new Titanic Belfast museum will be the world?s largest Titanic visitor attraction.

This befits a resurgent Belfast that was nominated by National Geographic Traveler magazine as one of the world?s top tourist attractions for 2012, and with the steady improvement of political relationships, the city looks set for much happier times.

PTS: US Olympic?dreams in grave danger

Building an Olympic team is tricky business. Because you?re constructing a team more or less from scratch, and focusing all efforts into a very limited number of meaningful games.

By the time any major flaws have been identified, it might be too late. Because flaws proving more significant than originally believed can turn fatal, and right quick.

There?s no sugar-coating it: the United States is in a dark corner after Saturday?s surprising 2-0 loss to Canada. Caleb Porter?s team must win Monday against El Salvador to ensure passage into the critical semifinals in Kansas City.

A draw against El Salvador, a tiny nation now inspired by the massive Olympic opportunity in front of them, probably won?t be enough for the U.S. under-23s.

So, about those ?flaws:?

Dynamic formations and shrewd, fluid arrangements of so much attacking talent doesn?t mean a thing, as we can see, if the old-fashioned elements of leadership and want-to come up missing. Or, perhaps, were never there in the first place. Without the benefit of meaningful matches to test these elements over the last few months well, you never really know until you know. You know?

Saturday, ?urgency? went on holiday on the U.S. side. The Canadians ran and ran and put everything they had into Saturday?s contest. Too many American players might be asking if they did the same Saturday.

Going into the match, you could say there was definitely still a little bolt-tightening to be done in the U.S. defense. Seeing things unravel so spectacularly against Canada, it looks now like more than that. The problems start with center back Ike Opara, who just never looks completely comfortable or very smooth back there. His timing and positioning aren?t as astute as central partner Perry Kitchen, but that?s not the least of it. Some nervous indecision near goal nearly turned disastrous as Opara almost sneaked one past Bill Hamid at the near post in the first half. ?He got all turned around on a Canadian break midway through the second half, and then completely lost his mark on Canada?s second goal.

Opara can definitely can be a bother on offensive set-plays. But that?s not enough, and that spot looks like a real doozey of a U.S. problem.

The first Canadian strike was clearly on goalkeeper Bill Hamid, whose inexperience became a crusher. He was way too timid in claiming what should have been a routine ball into his six-yard box. By failing to grab the floating corner kick at its highest possible point (Goalkeeping 101) and not attacking the moment with authority, he turned a fairly benign cross into a fiasco.

Freddy Adu isn?t playing badly, really, but he?s sure not anything to shout about, either. Bottom line: he?s not doing enough. The U.S. captain (and most experienced international man) must press the game more and ask further questions of defenders in front of him. He?s quite competent in helping the Americans keep possession in the middle third. But he?s not stretching defenders the way Brek Shea did on the left (for a half Saturday, anyway). And when Adu came into the middle after the break, he was even less effective, never establishing himself as the playmaker Porter apparently asked him to be in a halftime tactical adjustment.

Speaking of that tactical adjustment: it didn?t work. Not at all. Joe Corona, Thursday?s three-goal scorer, wasn?t finding the spaces that he did against Cuba?s awful defense. And, as noted, Adu wasn?t having his best night on the right wing. So Porter removed Corona, switched Shea to the right, redeployed Adu to attacking midfielder and added Joe Gyau to the left. Result: things got worse. Only in the last, desperate 10 minutes did the Americans begin seriously threatening Canadian goal. Down by two at that point, it was too late.

Zynga founder, other insiders to sell 43M shares

(AP) ? Zynga's CEO and other insiders at the online games company plan to sell 43 million shares of stock in a public offering that will boost the amount of stock available for general trading by 35 percent.

Class B stock owned by Mark Pincus and others will be converted to regular, Class A shares. All shares have the same ownership value, but Class B stock has seven times the voting power.

Pincus, who founded Zynga Inc. in 2007, will control about 35 percent of the company once the offering is complete. That's down only slightly from 36.5 percent because he will retain his Class C shares, which have 70 times the voting power of Class A stock.

Zynga, which had its initial public offering of stock in December, disclosed the latest plans in a regulatory filing Friday and said proceeds from the sale will go to the individuals, not the company.

Zynga is the maker of online games such as "CityVille," ''FarmVille and "Zynga Poker." Many of the games are played on Facebook, though Zynga has been working on expanding to other areas, notably mobile devices. "Words With Friends," a Scrabble-like game played mainly on smartphones, is one of its best-known titles.

It was one in a group of social Web companies that went public over the past year. Others include LinkedIn Corp., Groupon Inc. and Yelp Inc. Facebook is expected to have its IPO this spring.

The move announced Friday was expected. Last week, the company said Zynga shareholders may sell some shares, just three months after the company went public, to try to avoid a drop in its stock price. The number of shares wasn't disclosed at the time.

Early investors typically must wait about six months after an IPO to sell off parts of their stakes. By selling some shares now, shareholders won't be flooding the market all at once in June when the post-IPO "lock-up" is scheduled to expire. A mass wave of selling can drive down the stock price.

Zynga's stock fell 36 cents, or 2.6 percent, to close Friday at $13.40. At that price, the 43 million shares will be worth about $576 million. Pincus is offering 16.5 million shares, worth about $221 million. The stock has increased 34 percent since its IPO price of $10.

Zynga also disclosed that it had spent $180 million to buy OMGPop, maker of the popular mobile game "Draw Something." The company didn't disclose the price when it announced the deal on Wednesday.

Tax Cap Slows Pemex Finance | The Sharpener

MEXICO CITY ? Petroleos Mexicanos (Pemex) reported a loss of 88.400 billion pesos for the cap imposed by the Ministry of Finance ?when deducting production costs, labor or raw material.

This impact is greater than that of labor liabilities or LP gas subsidy for 2011.

Autism risk gene linked to differences in brain structure

ScienceDaily (Mar. 21, 2012) ? Healthy individuals who carry a gene variation linked to an increased risk of autism have structural differences in their brains that may help explain how the gene affects brain function and increases vulnerability for autism. The results of this innovative brain imaging study are described in an article in the groundbreaking neuroscience journal Brain Connectivity, a bimonthly peer-reviewed publication from Mary Ann Liebert, Inc. The article is available free online at the Brain Connectivity website.

"This is one of the first papers demonstrating a linkage between a particular gene variant and changes in brain structure and connectivity in carriers of that gene," says Christopher Pawela, PhD, Co-Editor-in-Chief and Assistant Professor, Medical College of Wisconsin. "This work could lead to the creation of an exciting new line of research investigating the impact of genetics on communication between brain regions."

Although carriers of the common gene variant CNTNAP2 -- identified as an autism risk gene -- may not develop autism, there is evidence of differences in brain structure that may affect connections and signaling between brain regions. These disruptions in brain connectivity can give rise to functional abnormalities characteristic of neuropsychological disorders such as autism.

Emily Dennis and coauthors from UCLA School of Medicine and UCLA (Los Angeles, CA) and University of Queensland and Queensland Institute of Medical Research (Brisbane, Australia), used a sophisticated imaging technique to study the brains of healthy young adults who are carriers of CNTNAP2.

Apple to pay dividend, start stock buybacks

NEW YORK (AP) ? Apple is finally acknowledging that it has more money than it needs. But don't expect it to cut prices on iPhones and iPads. Instead, the company said on Monday that it will reward its shareholders with a dividend and a stock buyback program.

Apple, the world's most valuable publicly traded company, sits on $97.6 billion in cash and securities.

The company has stockpiled the cash through a combination of great ideas and prudence. Apple spends money, to be sure, building data centers, buying parts for its products and pursuing ambitious projects such as a new 2.8-million-square-foot headquarters that has been likened to a spaceship. It also invests in the research and development of new technology and negotiates an occasional acquisition.

But Apple simply hasn't managed to spend its earnings faster than people are lining up to buy its iPads, iPhones and other gadgets.

The decision to return some of that money to investors is a clear signal that Apple is taking a different approach in the post-Jobs era.

Former CEO Steve Jobs resisted calls to issue dividends for years. He argued that the money was better used to give Apple maneuvering room to acquire other companies, for instance. Apple did pay a quarterly dividend between 1987 and 1995, but Jobs was not involved with the company at the time.

Jobs died in October after a long fight with cancer.

Since then, pressure had been mounting on new CEO Tim Cook. Apple's ever-growing pile of cash was earning a paltry amount of interest and the fact that it was sitting there unused could have left the company open to charges of mismanagement and possible shareholder lawsuits.

On Monday, Cook said that, with as much cash as Apple has on hand, a dividend won't restrain the company's options.

"These decisions will not close any doors for us," he told analysts and reporters on a conference call.

Indeed, Apple can afford it. The dividend, which should placate shareholders, will cost about $10 billion the first year. Apple generated $31 billion in cash in the fiscal year that ended in September and analysts expect it to add another $70 billion to $85 billion this year.

Apple said it will pay a quarterly dividend of $2.65 per share, starting in its fiscal fourth quarter, which begins July 1.

The dividend works out to $10.60 annually, or 1.8 percent of the current stock price. Although Microsoft Corp., pays 2.5 percent of its stock price in dividends, and Hewlett-Packard Co. pays 2 percent, analyst Tavis McCourt at Morgan Keegan said Apple's dividend is relatively generous for a large technology company.

Energy and phone companies often pay dividends worth more than 5 percent of their stock price.

In absolute terms, Apple will pay one of the richest dividends in the U.S. The roughly $10 billion it will spend in its first year, places it just below companies including AT&T Inc. and Verizon Communications Inc., which are among the biggest spenders because they use dividends as their main way to attract investors.

Exxon Mobil Corp., the world's second largest company by market capitalization, pays about $9 billion in dividends annually.

The dividend opens up ownership of Apple shares to a wider range of stock mutual funds, potentially boosting the stock price in the long term. Many "value-oriented" stock funds are not allowed to buy stocks that don't pay dividends.

Apple said the $10 billion share buyback program will begin next fiscal year, which starts Sept. 30, and runs for three years.

Investors had been expecting the announcement, driving Apple's stock up 37 percent since January, when management first hinted in that a dividend was in the works.

Buybacks are a popular alternative to dividends, since they reduce the number of shares outstanding. That means every remaining investor owns a larger share of the company.

Apple's stock hit a new high Monday before closing at $601.10, up $15.53. Since Steve Jobs' death on Oct. 5, Apple's stock is up nearly 60 percent. The company is worth $553 billion.

McCourt raised his price target on Apple's stock to $800 on Monday, becoming the first Wall Street analyst to do so. A dozen have price targets in the $700 range. He had been expecting the dividend, he said, and the main reason for the higher price target is the company's tremendous profit growth.

The dividend and buyback announcement comes three days after the launch of Apple's latest iPad tablet in the U.S. and nine other countries. Cook said sales the first few days set a record, but he gave no details.

Cook said the company also considered splitting its stock and continues to look at that option. Stock splits increase the number of shares while reducing their value, potentially making it easier for small investors to buy them. But Cook said "there's very little support" for the idea that stock splits can help the stock overall.

Cook suggested that the dividend could have been larger if U.S. tax laws were different.

Cook said that as Apple analyzed how much it could give out to shareholders, it looked solely at the cash it has in the U.S. Like many big exporters, Apple has much of its cash overseas ?some $64 billion, specifically.

Apple is reluctant to bring back overseas profits. In addition to being taxed in their respective countries, those profits would be subject to the 35 percent U.S. corporate tax rate.

"Current tax laws provide a considerable economic disincentive to U.S. companies that might otherwise repatriate a substantial amount of foreign cash," Chief Financial Officer Peter Oppenheimer said.

Cook said Apple looked at how much domestic cash it had, then set aside enough for planned investments and unforeseen outlays. What was left over would be given out to shareholders, he said.

That suggests that if Apple could bring back its $64 billion in overseas money, the rewards to shareholders could be larger. Corporations have been clamoring for a change in tax laws, or a repeat of a 2004 tax amnesty on repatriated earnings.
