miRNA tackles insulin-blocking protein

ScienceDaily (Oct. 12, 2012) ? Work by Michigan Technological University biologist Xiaoqing Tang is yielding new insights into how a tiny snippet of genetic material can promote healthy insulin production in mice.

Her work may eventually lead to new therapies for the treatment of diabetes, a disease that affects nearly 26 million Americans and causes myriad health problems, including heart disease, kidney failure and stroke. Diabetes results when the pancreas does not produce or release enough insulin into the blood stream or when cells fail to respond to the hormone.

The genetic material in question is a microRNA molecule called miR-30d, which is the same in mice and people. MicroRNA, or miRNA, attaches to long RNA molecules and prevents them from making proteins.

Proteins are the building blocks of life, but they can also cause serious problems; think of the plaques that develop in the brains of Alzheimer's patients.

One such protein is a tumor necrosis factor, which is involved in cell death and can trigger the production of another problematic protein, called MAP4K4, which blocks the formation of insulin when cells are under adverse conditions. MAP4K4 throws a wrench into the works by interfering with production of an important protein named MafA that binds to DNA and is an essential part of the insulin-making pathway.

In a series of experiments, Tang and her research team showed how miR-30d can counteract the tumor necrosis factor-triggered production of MAP4K4 and help the pancreas make more insulin.

First, they compared pancreas cells from diabetic mice with those of wild mice and found that the diabetic cells have much less miR-30d.

Second, using genes they created in their lab, they made cells that produce extra amounts of miR-30d. Those cells doubled the amount of the good protein MafA and generated much more insulin, showing that miR-30d works at least in part by activating MafA in the pancreas.

Finally, they added the tumor necrosis factor to those cells with the extra miR-30d. Unlike regular cells, which had MafA production blocked by the tumor necrosis factor, the super cells managed to keep on producing MafA, though not as much as before.

"What we found with miR-30d is that it can increase cells' ability to make insulin by activating MafA," Tang said. "We've also shown that the tumor necrosis factor-triggered MAP4K4 is a direct target of miR-30d. Based on our data, we think miR-30d probably plays multiple roles, both in enhancing insulin production and in protecting cells from the inflammatory effects of tumor necrosis factor."

Tang is now studying transgenic mice that generate extra amounts of miR-30d. "We want to induce diabetes and see if the process slows down in the transgenic mice," she said. "If that happened, it would be great."

The study is in its early stages, but preliminary results are intriguing. The transgenic mice are smaller and leaner than wild mice. Yet they don't seem to have extra insulin in their blood.

"We still don't understand why insulin is low in the blood of the transgenic mice." she said. "It may mean that insulin gets into cells from the blood very quickly. Or, the beta cells in the pancreas may sense that they don't need to produce much insulin. Or maybe it's another process all together. A mouse is much more complicated than a cell line."

Their latest research was published online Sept. 7 in The Journal of Biological Chemistry. The article, "MicroRNA-30d Induces Insulin Transcription Factor MafA and Insulin Production by Targeting Mitogen-Activated Protein 4 Kinase 4 in Pancreatic Beta Cells," was authored by Tang, Xiaomin Zhao and Ramkumar Mohan of Michigan Tech; and Sabire Ozcan of the University of Kentucky.

This is horser.net, a online social network for horse riders and everyone related to horses and riding. The website contains the full source code (developed in symfony) and a lot of domains, even trademarked Logo with a different name.

The script can be easily modified to run another social community like pet lovers (cats, dogs,...) or anything else, just name it.

The website has been developed from scratch within a timerange of about 2 years and can be extended with new modules / ideas if you can develop in symfony.

The website contains the following modules, already developed:

Community:

A complete community module with usual register / login process, own profile for each user, messaging a board, guestbook, Buddy-list, tagging, picture- and video galleries, groups and own blog for each user. Each user has it's own event calendar and can suggest events to the moderators, who then can put the event into a global calendar. A newsstream like at facebook shows, what's happeing with the latest activities. A member overview is also included.

Magazine:

Different editors can create articles and stories. They can be created within different categories. Each article creates automatically a shortened tweet and there can be also a video connected to an article.

CMS:

The page contais a special and very easy to use CMS module (we call it easyCMS). The editor can click his page together from different boxes like a title, text, text with a picture (left, middle, right), pictures, a form builder and a link box. Very easy to use, just drag n drop your article together.

The CMS contains also a great asset manager for uploading and managing all the media.

Video / Article dictionary:

We created a dictionary, kinda mini wiki for the page. You can insert videoclips or just text with pictures or a combination of everyting. Chose from different categories to tag them.

Horse marketplace:

We build a complete horse marketplace, where users could sell their horses very easily. You can create a picture gallery, show important infos about the horse, you can show a complete pedigree of the horse and also the necessary contact detaisl with a google map. Also the marketplace shows random advertisements from the marketplace offers on the sidebar if the seller has more horses to sell, also it shows a nice advertisement with logo from the seller there.

Global Marketplace:

Besides the marketplace for horses, we created also a global marketplace for all user, so they can sell there new and used goods to anyone. Products also contain picture gallery with slider, description, contact details, categories and so on.

B2B Yellow pages / trade directory:

The system includes a nice B2B directory which allows a user to register as a normal user or as a business user. B2B user can be charged for this service, so they can present their company within the portal. Every company get's it's own page with picture slideshow, logo, contact details, description, google map, description and so on. Companies will be divided into different categories, so it even more easier to find it. The is also a overview page for each category, showing a pic and the name. Also there is a nice detailed search incl. radius search as every adress, which is given at the registration process, beeing automatically geo-coded for google map.

Advertisement:

There are two banner spots, which are hardcoded atm with two graphics. Can easily exchanged by a advertisment code snippet. Also the system itself shows latest offers from marketplace and horse marketplace sitewide.

Start page:

The startpage contains a nice slider pointing to articles, videos or anything else (created by a easy xml file). You can login or register, you see the latest news and also main topic articles.

Other:

We put a lot of effort, brain and developing into this. The design is seperated from the code, so it's not so hard to create a new layout for the system. it is prepared for multilanguage, we didn't finish that completely, but not so much left for that.

Via the backend you can create and manage users and upload videos, create categories and stuff. Content will be mostly created as inline editing directly on the frontpage, so very easy to use.

The website originally started with the german name PferdeABC.com and later changed to horser.net.

Long-distance relationships aren?t easy. Especially when it comes to business.

You can also utilize productivity tools such as Google Docs and WebEx to make your virtual meetings more seamless by meeting online via mobile devices, sharing destkops, files and more.

2. Share tasks virtually.

Whenever we have a series of projects with multiple action items, I?ll set up a shared to-do list online. There are numerous task and project-oriented apps on the market that make collaboration easier for small businesses such as Asana, HiTask and others.

The entire team works closely on projects, so it?s a great way to share responsibilities without bulky software or endless emails about who is doing what at any given time.
